Sacituzumab tirumotecan improved progression-free and overall survival compared with chemotherapy in patients with advanced or recurrent endometrial cancer previously treated with platinum-based chemotherapy and immunotherapy.

What TroFuse-005 found

The randomized phase 3 trial enrolled 776 patients. Median overall survival was 19.0 months with sacituzumab tirumotecan and 12.1 months with physician-selected chemotherapy. Median progression-free survival was 7.2 versus 3.9 months.

At the prespecified analysis, the hazard ratio for death was 0.68. That describes the relative hazard over follow-up, not a guarantee of how long an individual patient will live.

The safety findings belong beside the benefit

Grade 3 or higher adverse events occurred in 69% of treated patients receiving the study drug and 57% receiving chemotherapy. Treatment-related deaths occurred in three patients in each group.

The trial was open-label, although central radiology review was blinded, and was funded by Merck Sharp & Dohme. The results were published on 26 October. They do not, by themselves, establish regulatory approval, affordability or availability in South Africa.

InsideHer’s editorial view is that a cancer-treatment story should give survival, toxicity and access equal attention. The result concerns a defined treatment setting and should not be generalised to all endometrial cancer.

Regulatory decisions, longer follow-up, quality-of-life results, treatment access and comparative evidence.
